Although there are more and more new drug molecules developed to fight antimicrobial resistance or life-threatening diseases such as cancer, many of them have not made it yet into clinical trials or the market because their interaction with biological membranes is poorly understood. In my PhD project I use a class of polymeric molecules for drug delivery with a defined structures called dendrimers to study their interactions with a number of lipid membrane models of varying complexity. I am using a various biophysical methods, such as scattering (neutrons, X-rays, DLS), spectroscopic (UV-Vis, FTIR, Fluorescence), and thermal (DSC, nano-DSC) techniques and surface pressure measurements to probe the influence of surface charge, solution salinity, pH, temperature or methanol content on the interaction of the dendrimer molecules with the lipid membranes but also on the behaviour of the molecules on their own in solution. During the biophysical testing we also had access to the INTER and SURF beamline at the ISIS neutron source and the BioSAXS B21 beamline at Diamond Light Source (Harwell, UK). In collaboration with the Maths Department there is another PhD student working on simulations and theoretical modelling on this project to match my experimental results. More complex studies involve cytotoxicity testing on a variety of cancer cell lines and antimicrobial activity screening on bacteria and fungi.
